*Thesis Abstract:*

*Vitex negundo, locally known as lagundi, is a folkloric medicinal plant
that is endemic in the Southeast Asian region. It is now used to treat a
variety of conditions such as asthma and cough. There is need for a rapid
analysis of its extract in establishing a quality control parameter in the
field. In this work, thin layer chromatography (TLC) is coupled with
digital imaging technology to characterize the methanol extract of V.
negundo – one obtains additional data from the TLC image data such as
color, intensity and retention factor (Rf) versus Rf data only in
traditional TLC runs. The chromatograms were processed using ImageJ®
software wherein the TLC bands were converted into plots that serve as the
plant sample profile. TLC runs at different time frames between 5 to 30
minutes were compared. For each time frame, color separation using RGB
stack analysis was used to characterize the bands. These were also compared
with the standard agnuside, a molecular marker present in lagundi extract.
Adulterated samples (mixed with Blumea balsamifera) and degraded leaf
samples (heated at 80OC or exposed to moisture for 24 h) were also
extracted and compared with good samples of lagundi. The results
established a proof-of-concept demonstration that rapid TLC and image plot
fingerprinting to qualify good and bad plant samples.*
